神経回路の可塑性の方向性,つまりシナプス結合の増強・抑圧の方向性を定めるメカニズムとして spike-timing-dependent plasticity (STDP, 発火時間に依存する可塑性) が注目されている. 近年の単一細胞を用いた生理実験から,興奮性シナプス後電位 (Excitatory Post-synaptic Potential, EPSP) が発火電位 (Action Potential) に先行するとシナプス結合は増強され,その逆の場合に抑圧されることが示されている. 本報告では,この STDP を応用し,微小電気刺激や経頭蓋磁気刺激 (TMS) を用いて脳の神経回路を再構築する試みについて解説する. 外部刺激で誘発される皮質内の EPSP と,電気刺激や TMS で誘発される AP との時間差を正確に制御することで,皮質の特定の回路にSTDPを誘発できる.例えば,ある特定の外部刺激に対応する投射経路を抑圧し,異なる投射経路を増強することで,皮質の神経回路を”書き換え”,外部刺激に対する反応を改変できる. 筆者らの研究グループではラット聴皮質において,音刺激と微小電気刺激のタイミングを調整することで,任意の周波数に対する反応を増強・抑圧することに成功した. また,他のグループによる研究からは,ヒトの視覚野で誘発された STDP が,視覚認知をも改変しうることがわかっている.
